Asymmetric distribution of irreversibly oxidized, ClpB-associated proteins in stressed mycobacteria.
"Mycobacterium tuberculosis (Mtb) co-evolved with humans to withstand stresses imposed by the immune system and uses similar defenses against chemotherapy. Besides catabolizing potentially bactericidal compounds, Mtb can repair damaged molecules or degrade them. We used new chemical and biochemical probes and improved image analysis algorithms for time-lapse microscopy to reveal an additional line of defense of mycobacteria against stationary phase stress, oxidants and antibiotics— the ClpB-associated sequestration of irreversibly oxidized proteins (IOP) into aggregates, followed by the asymmetric distribution of aggregates within bacteria and between their progeny. Progeny born with minimal IOP grew faster and better survived a subsequent antibiotic stress than their IOP-burdened sibs. ClpB-deficient Mtb had a marked recovery defect from stationary phase or antibiotic exposure and survived poorly in mice. Treatment of tuberculosis might be assisted by drugs that cripple the pathway by which Mtb buffers, sequesters and asymmetrically distributes IOP. "

"Alzheimer's disease and the related diseases, which are part of the everyday routine of the memory consultation nurses, are only known by clinicians and the general public when the patients and/or their next of kin request treatment. What happens before this? It is essential that we find out more about this because this is probably when preventive or curative treatment can actually have an effect. When dealing with a sickness that is dominated by a loss of memory and self-awareness, you cannot rely on the answers given to questions about the past. Tracking the illness within the population well before treatment is required is therefore the only way of understanding its hidden phase. This is what we have been doing with Paquid, 3C and AMI for the last 30 years. We now know what happens 20 years before the need for treatment, and Prof Dartigues will talk to us about this…"
